Gramocef Injection is available in strengths containing Ceftriaxone 250 mg, 500 mg, 1 g, and 2 g for intravenous (IV) or intramuscular (IM) administration.

Gramocef Injection contains Ceftriaxone, a third-generation cephalosporin antibiotic with broad-spectrum bactericidal activity against a wide range of Gram-positive and Gram-negative bacteria. It offers excellent tissue penetration and prolonged duration of action, allowing convenient once-daily dosing in many clinical situations. Ceftriaxone remains active against numerous β-lactamase-producing organisms, making it an effective treatment option for severe and complicated bacterial infections.

Gramocef Injection is indicated for the treatment of urinary tract infections, lower respiratory tract infections, bacteremia, septicemia, meningitis, intra-abdominal infections, and infections caused by susceptible Pseudomonas species. It is also used for surgical prophylaxis and the treatment of uncomplicated gonococcal infections.

Ceftriaxone is a bactericidal cephalosporin antibiotic that acts by inhibiting bacterial cell wall synthesis. It binds to specific penicillin-binding proteins (PBPs), disrupting the final stages of peptidoglycan synthesis and leading to bacterial cell death. Ceftriaxone demonstrates stability against many β-lactamases, including certain penicillinases and cephalosporinases produced by Gram-positive and Gram-negative organisms, thereby maintaining activity against a broad spectrum of susceptible pathogens.

The usual adult dosage is 1–2 g administered once daily or in equally divided doses depending on the severity and type of infection. The total daily dose should not exceed 4 g. For uncomplicated gonococcal infections, a single intramuscular dose of 250 mg is recommended. For surgical prophylaxis, a single intravenous dose of 1 g should be administered 30 minutes to 2 hours before surgery. Dosage adjustments should be made according to the patient's clinical condition and physician's recommendation.

Gramocef Injection is contraindicated in patients with known hypersensitivity to Ceftriaxone, cephalosporins, or any component of the formulation. It is also contraindicated in neonates requiring treatment with calcium-containing intravenous solutions, including parenteral nutrition, due to the risk of ceftriaxone-calcium precipitation. Hyperbilirubinemic neonates, particularly premature infants, should not receive Ceftriaxone therapy.
