Composition
Each film-coated tablet contains Dapagliflozin Propanediol Monohydrate equivalent to Dapagliflozin 10 mg, Linagliptin 5 mg, and Metformin Hydrochloride Sustained Release 500 mg or 1000 mg.

Dapagliflozin + Linagliptin + Metformin SR Tablets combine three complementary antidiabetic agents to provide comprehensive glycemic control in adults with type 2 diabetes mellitus. Dapagliflozin, an SGLT2 inhibitor, lowers blood glucose by promoting urinary glucose excretion. Linagliptin, a DPP-4 inhibitor, increases endogenous incretin hormone levels, thereby enhancing glucose-dependent insulin secretion and reducing glucagon release. Metformin, a biguanide, decreases hepatic glucose production, improves insulin sensitivity, and reduces intestinal glucose absorption. The sustained-release formulation of Metformin allows convenient once-daily dosing while supporting improved treatment adherence and long-term glycemic management.
Indications
Dapagliflozin + Linagliptin + Metformin SR Tablets are indicated as an adjunct to diet and exercise for improving glycemic control in adults with type 2 diabetes mellitus.
Pharmacodynamics
Dapagliflozin is a selective sodium-glucose co-transporter 2 (SGLT2) inhibitor that reduces renal glucose reabsorption, resulting in increased urinary glucose excretion and improved glycemic control in patients with type 2 diabetes mellitus. Linagliptin is a reversible dipeptidyl peptidase-4 (DPP-4) inhibitor that increases circulating incretin hormone concentrations, thereby enhancing glucose-dependent insulin secretion and suppressing glucagon release. Metformin Hydrochloride decreases hepatic glucose production, reduces intestinal glucose absorption, and improves peripheral insulin sensitivity. Clinical studies have also demonstrated that Metformin is associated with weight neutrality or modest weight loss. Together, these complementary mechanisms provide effective and sustained blood glucose control.
Dosage and administration
The recommended dosage is one tablet taken orally once daily, with or without food, or as directed by the treating physician. The tablet should be swallowed whole and must not be crushed, broken, or chewed before swallowing. Dosage should be individualized according to the patient's clinical response, renal function, and glycemic control.
Contraindications
This combination is contraindicated in patients with a history of serious hypersensitivity reactions to Dapagliflozin, Linagliptin, Metformin, or any component of the formulation, including anaphylactic reactions, angioedema, urticaria, or bronchial hyperreactivity. It is also contraindicated in patients with severe renal impairment (eGFR below 30 mL/min/1.73 m²), end-stage renal disease (ESRD), or patients undergoing dialysis.
