Each uncoated bilayered tablet contains Pioglitazone Hydrochloride IP equivalent to Pioglitazone 15 mg + Glibenclamide IP 5 mg + Metformin Hydrochloride IP 500 mg (Sustained Release).

Pionorm-GM Tablets combine three complementary antidiabetic agents—Pioglitazone, Glibenclamide, and Metformin—to provide comprehensive glycaemic control in patients with type 2 diabetes mellitus inadequately controlled on dual therapy. Pioglitazone improves insulin sensitivity, glibenclamide stimulates pancreatic insulin secretion, and metformin reduces hepatic glucose production while improving peripheral glucose utilization. This triple combination targets multiple pathophysiological defects of type 2 diabetes, helping achieve better long-term glycaemic control.

Pionorm-GM Tablets are indicated as a third-line treatment for adults with type 2 diabetes mellitus when diet, exercise, monotherapy, or dual-drug therapy fails to achieve adequate glycaemic control.

Pioglitazone is a thiazolidinedione that decreases insulin resistance in peripheral tissues and the liver, resulting in increased insulin-dependent glucose disposal and reduced hepatic glucose output. Metformin is an antihyperglycaemic agent that lowers both basal and postprandial plasma glucose by decreasing hepatic glucose production, reducing intestinal glucose absorption, and improving peripheral insulin sensitivity. Glibenclamide, a second-generation sulfonylurea, stimulates insulin secretion from functional pancreatic beta cells, thereby promoting glucose utilization and lowering blood glucose levels. The combination provides complementary mechanisms for effective glycaemic management.

The recommended dosage is one tablet once daily with a meal, or as directed by the physician. Dosage should be individualized according to the patient's glycaemic response and clinical condition.

Pionorm-GM Tablets are contraindicated in patients with hypersensitivity to pioglitazone, glibenclamide, metformin, or any component of the formulation; cardiac failure or a history of cardiac failure (NYHA Class I–IV); hepatic impairment; diabetic ketoacidosis; active bladder cancer or a history of bladder cancer; unexplained haematuria; acute or chronic metabolic acidosis including diabetic ketoacidosis with or without coma; and type 1 diabetes mellitus.
