Composition
Each uncoated tablet contains Pioglitazone 15 mg/30 mg.
Features / Experimentation
Pionorm Tablets contain Pioglitazone, a thiazolidinedione (TZD) oral antidiabetic agent that improves insulin sensitivity in peripheral tissues and the liver. It enhances insulin-dependent glucose utilization, reduces hepatic glucose production, and improves glycaemic control without stimulating insulin secretion. As a selective PPAR-γ agonist, pioglitazone also positively influences glucose and lipid metabolism, making it suitable for patients with insulin resistance associated with type 2 diabetes mellitus.
Indications
Pionorm Tablets are indicated as an adjunct to diet and exercise to improve glycaemic control in adult patients with type 2 diabetes mellitus (Non-Insulin Dependent Diabetes Mellitus - NIDDM).
Pharmacodynamics
Pioglitazone is a thiazolidinedione antidiabetic agent whose activity depends on the presence of endogenous insulin. It reduces insulin resistance in peripheral tissues and the liver, resulting in increased insulin-mediated glucose uptake and decreased hepatic glucose production. Unlike sulfonylureas, pioglitazone does not stimulate insulin secretion. It acts as a potent and highly selective agonist of peroxisome proliferator-activated receptor-gamma (PPAR-γ), regulating the expression of insulin-responsive genes involved in glucose and lipid metabolism.
Dosage and administration
The recommended starting dose is 15 mg or 30 mg once daily. Depending on the patient's glycaemic response, the dose may be increased gradually up to a maximum of 45 mg once daily, as directed by the physician.
Contraindications
Pionorm Tablets are contraindicated in patients with known hypersensitivity to pioglitazone or any of the formulation components, cardiac failure or a history of cardiac failure (NYHA Class I–IV), hepatic impairment, diabetic ketoacidosis, active bladder cancer, history of bladder cancer, and patients with unexplained haematuria.
