Composition
Each tablet contains Sitagliptin 100 mg + Pioglitazone 15 mg + Metformin Hydrochloride SR 500 mg/1000 mg.
Features / Experimentation
SITAVIA-PM Tablets combine Sitagliptin (DPP-4 inhibitor), Pioglitazone (thiazolidinedione), and Sustained-Release Metformin (biguanide) to provide comprehensive glycaemic control in adults with type 2 diabetes mellitus. Sitagliptin enhances glucose-dependent insulin secretion by prolonging incretin activity, Pioglitazone improves insulin sensitivity in peripheral tissues and the liver, while Metformin reduces hepatic glucose production and enhances peripheral glucose utilization. The triple-drug combination targets multiple pathophysiological defects of type 2 diabetes for improved glycaemic management.
Indications
SITAVIA-PM Tablets are indicated as an adjunct to diet and exercise to improve glycaemic control in adults with type 2 diabetes mellitus.
Pharmacodynamics
Sitagliptin is a DPP-4 inhibitor that increases active incretin hormone concentrations, thereby enhancing glucose-dependent insulin secretion and reducing glucagon secretion. Pioglitazone is a thiazolidinedione that improves insulin sensitivity in peripheral tissues and the liver, resulting in increased insulin-dependent glucose disposal and reduced hepatic glucose production. Metformin is an antihyperglycaemic agent that decreases hepatic glucose production, reduces intestinal glucose absorption, and improves peripheral insulin sensitivity, lowering both fasting and postprandial plasma glucose levels.
Dosage and administration
The recommended dosage is one tablet orally once daily, with or without meals. The tablet should be swallowed whole or as directed by the physician.
Contraindications
SITAVIA-PM Tablets are contraindicated in patients with renal impairment, known hypersensitivity to sitagliptin, pioglitazone, metformin, or any component of the formulation, and in patients with acute or chronic metabolic acidosis, including diabetic ketoacidosis.
