Each tablet contains Trypsin BP 48 mg + Bromelain 90 mg + Rutoside Trihydrate 100 mg + Aceclofenac 100 mg.

Tolpazen is a multi-enzyme anti-inflammatory formulation combining Trypsin, Bromelain, Rutoside Trihydrate, and Aceclofenac to provide comprehensive management of pain, inflammation, edema, and tissue injury. The combination offers potent anti-inflammatory, analgesic, antioxidant, and anti-edematous effects, promoting faster recovery from musculoskeletal injuries and inflammatory disorders while reducing swelling and improving joint mobility.

Tolpazen is indicated for the treatment of inflammation associated with rheumatic and degenerative conditions, including rheumatoid arthritis, osteoarthritis, spondylopathies, tendinitis, and bursitis. It is also useful in painful inflammatory conditions where reduction of edema and improvement of tissue healing are desired.

Trypsin is a proteolytic enzyme that digests proteins into peptides and contributes to tissue repair by improving humoral immune response, inhibiting pathogen growth, reducing C-reactive protein levels, and exerting antioxidant effects through modulation of protease-activated receptor-2, thereby reducing inflammatory responses. Bromelain possesses potent anti-inflammatory properties by reducing neutrophil migration and suppressing the secretion of pro-inflammatory cytokines. Rutoside Trihydrate exhibits antioxidant and anti-inflammatory activity by inhibiting the transcription of pro-inflammatory genes in macrophages, thereby reducing vascular permeability and tissue inflammation. Aceclofenac is a phenylacetic acid derivative NSAID that provides analgesic and anti-inflammatory effects through preferential inhibition of cyclooxygenase-2 (COX-2), resulting in decreased prostaglandin synthesis and symptomatic relief from pain and inflammation.

The recommended dosage is one tablet once daily with or after food, or as directed by the treating physician.

Tolpazen is contraindicated in patients with known hypersensitivity to any of the active ingredients or excipients, severe congenital or acquired coagulation disorders, and in patients with a history of gastrointestinal bleeding or gastrointestinal perforation.
