Anti-arrhythmic formulation used for the management and control of ventricular and supraventricular arrhythmias. Helps stabilize cardiac rhythm and reduce abnormal electrical activity of the heart.

Indications

Indicated for the treatment and control of ventricular and supraventricular arrhythmias where other therapies are ineffective or unsuitable, including arrhythmias associated with Wolff-Parkinson-White syndrome.

Pharmacodynamics

Amiodarone exhibits anti-arrhythmic activity primarily by prolonging myocardial cell action potential duration and refractory period. It also exerts non-competitive antagonism of alpha- and beta-adrenoceptors, helping regulate abnormal cardiac conduction and rhythm disturbances.

Dosage and administration

Initial loading dose: 800–1600 mg/day until therapeutic response is achieved (usually within 1–3 weeks). Reduce to 600–800 mg/day for one month, followed by maintenance dose of approximately 400 mg/day or as directed by the physician.

Contraindications

Contraindicated in patients with cardiogenic shock, severe sinus node dysfunction causing marked sinus bradycardia, second- or third-degree atrioventricular block, syncope-associated bradycardia, corneal refractive laser surgery and hypersensitivity to amiodarone, iodine or formulation components.
