Features / Experimentation
Rivolas Tablets contain Rivaroxaban, a highly selective, orally active direct Factor Xa inhibitor that provides effective anticoagulation without requiring antithrombin III as a cofactor. By selectively inhibiting Factor Xa, Rivaroxaban reduces thrombin generation and thrombus formation while preserving predictable pharmacokinetics that eliminate the need for routine coagulation monitoring in most patients. Rivolas is indicated for the prevention and treatment of thromboembolic disorders across multiple cardiovascular conditions.
Indications
Rivolas Tablets are indicated for the prevention of atherothrombotic events in adult patients following acute coronary syndrome (ACS) when administered in combination with acetylsalicylic acid (ASA), with or without Clopidogrel or Ticlopidine. They are also indicated for the prevention of venous thromboembolism (VTE) following elective hip or knee replacement surgery, treatment of deep vein thrombosis (DVT) and pulmonary embolism (PE), prevention of recurrent DVT and PE, and prevention of stroke and systemic embolism in patients with non-valvular atrial fibrillation.
Pharmacodynamics
Rivaroxaban is a potent, selective, direct Factor Xa inhibitor that blocks the active site of Factor Xa without requiring antithrombin III for its anticoagulant activity. Factor Xa occupies a central position in the coagulation cascade by converting prothrombin to thrombin through both intrinsic and extrinsic pathways. Inhibition of Factor Xa effectively reduces thrombin generation, fibrin clot formation, and thrombus development, thereby lowering the risk of arterial and venous thromboembolic events.
Dosage and administration
For non-valvular atrial fibrillation, the recommended dosage is 15 mg or 20 mg once daily with food, depending on renal function. For the treatment of deep vein thrombosis (DVT) and pulmonary embolism (PE), the recommended dosage is 15 mg twice daily with food for the first 21 days, followed by 20 mg once daily with food. For reducing the risk of recurrent DVT and PE after at least six months of standard anticoagulant therapy, the recommended dosage is 10 mg once daily with or without food. For prophylaxis of DVT following hip or knee replacement surgery, the recommended dose is 10 mg once daily with or without food. For prophylaxis of venous thromboembolism in acutely ill medical patients at risk of thromboembolic complications, the recommended dose is 10 mg once daily for a total duration of 31 to 39 days. For patients with acute coronary syndrome (ACS), the recommended dosage is 2.5 mg twice daily in combination with Aspirin 75–100 mg once daily, with or without Clopidogrel or Ticlopidine, as directed by the physician.
Contraindications
Rivolas Tablets are contraindicated in patients with active pathological bleeding, severe hypersensitivity to Rivaroxaban or any component of the formulation, and severe renal insufficiency. The product should also be avoided in patients for whom anticoagulant therapy is contraindicated due to active bleeding disorders.
